Researchers find immunity protein that ramps up inflammation, and agents that can block it

Mar. 31, 2013 ? Scientists at the University of Pittsburgh School of Medicine have discovered a new biological pathway of innate immunity that ramps up inflammation and then identified agents that can block it, leading to increased survival and improved lung function in animal models of pneumonia.

They reported their findings today in Nature Immunology.

Pneumonia and other infections sometimes provoke an inflammatory response from the body that is more detrimental than the disease-causing bacteria, said senior author Rama Mallampalli, M.D, professor and vice chair for research, Department of Medicine, and director of the Acute Lung Injury Center of Excellence at Pitt.

"In our ongoing studies of pneumonia, we found infecting bacteria activate a previously unknown protein called Fbxo3 to form a complex that degrades another protein called Fbxl2, which is needed to suppress the inflammatory response," said Dr. Mallampalli, who is also chief of the pulmonary division of the VA Pittsburgh Healthcare System. "The result is an exaggerated inflammatory response that can lead to further damage of the lung tissue, multi-organ failure and shock."

The research team, led by Bill B. Chen, Ph.D., associate professor, Division of Pulmonary, Allergy and Critical Care Medicine, conducted experiments in which mice that lacked the ability to make Fbxo3 were infected with a strain of Pseudomonas bacteria, and found that they had better lung mechanics and longer survival than mice that still made the protein.

Research team members Bryan J. McVerry, M.D., and Yingze Zhang, Ph.D., both of the Acute Lung Injury Center of Excellence, found that blood samples from 16 people who had sepsis, a condition of systemic inflammation, revealed higher levels of Fbxo3 and other inflammatory proteins and lower levels of Fbxl2 than samples from seven patients who did not have sepsis or lung infection.

Based on the structure of Fbxo3, the researchers developed a family of small molecules with the aim of inhibiting its activity. Administration of one of them, called BC-1215, led to reduced inflammatory markers and improved lung mechanics in mouse models of pneumonia and sepsis.

"The key is to find ways to help the body temper its inflammatory response so that it's able to kill the infectious agent without causing injury to healthy tissue," Dr. Mallampalli said.

"The F-box protein Fbxo3, and other related proteins, represent ideal targets for treatment of acute lung injury, because it controls the innate immune response, is upstream of important inflammatory signaling pathways, and is more selective than traditional drugs that regulate protein turnover," noted Mark T. Gladwin, M.D., chief of the Division of Pulmonary, Allergy and Critical Care Medicine, Pitt School of Medicine.

The team is beginning to study the effects of BC-125 on other conditions of systemic inflammation, such as colitis and arthritis.

Co-authors of the paper include Tiffany A. Coon, Jennifer R. Glasser, Jing Zhao, Ph.D., Yutong Zhao, M.D, Ph.D., Chunbin Zou, Ph.D, Bryon Ellis, and Frank C. Sciurba, M.D., all of the Acute Lung Injury Center of Excellence, University of Pittsburgh.

(AP) ? Police said Saturday they are looking for a transient in the kidnapping of a 10-year-old girl who was snatched from her San Fernando Valley home before dawn last week and abandoned hours later in front of a hospital.

Investigators identified 30-year-old Tobias Dustin Summers as a suspect in the case but couldn't elaborate on the motive or what led them to him. Police don't know if the girl was targeted but said they don't believe Summers had a connection to her family.

"We have no information that the family knew this individual or that the individual knew any members of the family," Los Angeles Police Deputy Chief Kirk Albanese said.

About 40 detectives have been working around the clock looking for clues since the girl was abducted from her home Wednesday. She was found hours later, wandering near a Starbucks several miles away.

The girl was barefoot, had bruises and scratches, and wasn't wearing the same clothes she had on when she vanished. She told the police two men she didn't recognize had taken her from her home.

Police initially said they were looking for two suspects, but now are focusing their efforts on locating Summers.

"This is the only person we are looking for right now," Albanese said Saturday.

Investigators have said they believe the girl was driven around the San Fernando Valley in a couple of cars and taken to at least two locations, including a storage facility, before she was released.

A passer-by who recognized her picture from media reports saw her outside the Starbucks and called police. The girl had wandered there from the hospital where she had been dropped.

Summers, who has a distinctive tattoo of a ghoulish face on his right arm, has arrests dating back to 2002, police said. Among them are robbery, grand theft auto, possession of explosives and kidnapping, authorities said.

Police said they had no details on the prior kidnapping case.

Summers was released from prison in July on a petty theft conviction as part of a California law designed to ease crowding in state prisons. He also spent six days behind bars in January on a probation violation.

Summers last checked in with his probation officer at some point earlier this month and had been complying with his release terms, police said. He is known to frequent the area where the kidnapping took place.

The Los Angeles Times reported that law enforcement sources said the girl was sexually assaulted. The Associated Press does not identify victims of sexual assault. Summers isn't a registered sex offender, police said.

Albanese said Summers had been arrested four years ago for investigation of battery that involved child annoyance. Court records show Summers was convicted of battery in September 2009 but the child annoyance charge was either dismissed or not prosecuted.

Summers has family in Southern California, according to police, and the FBI said it will obtain a warrant for unlawful flight to avoid prosecution, if the agency determines he has fled the state.

NYC suspect in shooting of 4 released from custody

NEW YORK (AP) ? The man arrested in the fatal shooting of one person and wounding three of others in Brooklyn's Coney Island neighborhood earlier this month has been released from custody.

A spokesman for the Brooklyn District Attorney's office says Saturday a judge ordered 29-year-old Joseph Brown released on Friday.

The spokesman says the investigation is ongoing and Brown is due back in court in May.

Police had said Brown entered an apartment in a public housing complex, walked to a back bedroom and shot a man in the head. Investigators said he then shot three other people in the apartment, killing one.

We know the OUYA game console began shipping out to the Kickstarter backers as of yesterday, however we are now seeing some additional details. These are details in terms of the shipping process as well as the upcoming retail launch date. The folks at OUYA have also given an updated status in terms of the amount of available games and left things with a promise that these shipments are ?only the beginning.?

Mate choice in mice is heavily influenced by paternal cues, mouse study shows

Mar. 28, 2013 ? Hybrid offspring of different house mice populations show a preference for mating with individuals from their father's original population.

Mate choice is a key factor in the evolution of new animal species. The choice of a specific mate can decisively influence the evolutionary development of a species. In mice, the attractiveness of a potential mate is conveyed by scent cues and ultrasonic vocalizations. Researchers from the Max Planck Institute for Evolutionary Biology in Pl?n investigated whether house mice (Mus musculus) would mate with each other even if they were from two populations which had been separated from each other for a long time period. To do this, the researchers brought together mice from a German population and mice from a French population. Although to begin with all the mice mated with one another randomly, the hybrid offspring of French and German parents were distinctly more choosy: they showed a definite preference for mating with individuals from their father's original population. According to the researchers, this paternal imprinting accelerates the divergence of two house mouse populations and thus promotes speciation.

In allopatric speciation, individuals of a species become geographically isolated from each other by external factors such as mountains or estuaries. Over time, this geographic separation leads to the sub-populations undergoing various mutations, and thus diverging genetically. Animals from the two different sub-populations can no longer successfully reproduce, so two new species evolve.

To find out what role partner selection plays in such speciation processes, Diethard Tautz from the Max Planck Institutefor Evolutionary Biology and his colleagues conducted a comprehensive study on house mice -- the classic model organisms of biology. "To investigate whether there are differences in the mating behaviour of the mice in the early stages of speciation, we caught wild house mice in southern France and western Germany. The two populations have been geographically separate for around 3,000 years, which equates to some 18,000 generations," says Diethard Tautz. Due to this geographical separation, the French and German mice were genetically different.

The Pl?n-based researchers created a semi-natural environment for their investigations -- a sort of "Playboy Mansion" for mice. The research enclosure was several square meters in size and was divided up using wooden walls, "nests" made out of plastic cylinders, and plastic tubes. It also featured an escape tube with several entrances, which led into a cage system nearby. "We constructed the enclosure in such a way that all animals had unimpeded access to all areas, but thanks to the structural divisions were also able to create their own territories or retreat into nests," explains Tautz. "The escape tube was a control element. If the mice retreated to it only very seldom -- as was the case in our experiment -- then we could be sure there was no overpopulation in the central enclosure."

In this central enclosure, the French and German mice had both time and space to mate with each other and reproduce. "At first, all the mice mated with each other quite randomly. But with the first-generation offspring, a surprising pattern emerged," says Tautz. When the first-generation hybrid offspring of mixed French and German parentage mated, they showed a specific preference for pure-bred mates whose "nationality" was that of their father only. "There must be some kind of paternal influence that prompts the hybrid mice to choose a mate from a specific population, namely that of their father," concludes the biologist, based on the results of his study. "This imprinting must be learned, however, meaning that the animals must grow up in the presence of their fathers. This was not the case for the original mice, which were kept in cages for a time after being caught."

"We know that mice use ultrasonic vocalizations to communicate with each other and that particularly in the case of male mice these vocalizations can reveal signals of individuality and kinship. We believe that, like birdsong, the vocalizations of the males have a learned component and a genetic component," says Tautz. Therefore, French and German mice really could "speak" different languages, partly learned from their fathers, partly inherited from them. Individual mice thus have a mating preference for mice that speak the same language as they do.

The French and German mouse populations had evidently been geographically separated long enough for preliminary signs of species differentiation to be apparent as regards mating preferences. In addition, another aspect of mating behavior also sped up the speciation process. Although mice have multiple mates, the researchers found evidence of partner fidelity and inbreeding. The tendency to mate with relatives fosters the creation of genetically uniform groups. When both occur together, this accelerates the speciation process.

In a next step, Diethard Tautz wants to find out whether the vocalizations of the mice play the decisive role in paternal imprinting, or if scent cues are also involved. Furthermore, the biologist wants to identify the genes that are involved in mate selection.

GameStick is here ... er, sort of. The development units are at least here at GDC 2013, quietly hanging out behind a nondescript computer monitor running a few Android games paired with a Nyko wireless controller. That's right, it wasn't running with the proprietary GameStick controller, but a third-party wireless -- we'll go hands-on with the company's internally built controller later this evening, and thusly we didn't want to offer a half-impression with the third-party units available on the show floor. Stay tuned for more, and take a gander at the gallery below for now.

The Truth Behind the 'Biggest Cyberattack in History'

Is it "the biggest cyberattack in history"? Or just routine flak that network-security providers face all the time?

News websites across the Western world proclaimed Internet Armageddon today (March 27), largely due to a New York Times story detailing a "squabble" between the spam-fighting vigilantes at Spamhaus and the dodgy Dutch Web-hosting company Cyberbunker.

"Fight Jams Internet," the Times headline said. "Global Internet slows," the BBC proclaimed in the wake of the Times' story. Both websites alleged that Netflix streaming was slowing down as a result.

The reality is less exciting, though still serious. The Internet disruptions, which were centered in Western Europe, appear to be largely over, and were largely unnoticed even when occurring.

But, if anything, the incident may prompt a fix for a basic security flaw in the Domain Name System that serves as one of the underpinnings of the Internet.

"Despite the work that has gone into making the Internet extremely resilient, these attacks underscore the fact that there are still some aspects of it that are relatively fragile," said Andrew Storms, director of security operations at San Francisco-based network-security provider nCircle.

Too much information

Cyberbunker appears to be behind a massive distributed denial-of-service (DDoS) attack that first tried to first take down Spamhaus, then Spamhaus' network-reliability provider CloudFlare, and finally this past Saturday (March 23) hit CloudFlare's own bandwidth providers in Europe.

Boston-based Akamai Networks told the Times, and Spamhaus told the BBC, that the last round of attacks peaked at 300 gigabits per second, possibly the largest amount of bandwidth ever recorded during a DDoS attack.

According to a CloudFlare blog posting, the attack was launched on March 18 and immediately involved a tactic called DNS amplification, in which unprotected Domain Name System (DNS) servers are used to flood targeted servers with huge amounts of useless information, tying up bandwidth and processing time.

The attacks increased in volume during the week, finally peaking on Saturday when, according to CloudFlare, half of the infrastructure on the London Internet Exchange, an Internet node connecting several large-scale networks, was tied up by the attack. (CloudFlare is based in Palo Alto, Calif., but runs a global network.)

DNS servers are essentially the phone books of the Internet. Every Internet-connected device, from your computer to your smartphone, uses them to match a website address that humans use, such as "www.technewsdaily.com," with an Internet Protocol address that computers and routers use, such as "207.86.128.60."

DNS servers are essential, yet many remain "open," which means they will accept lookup requests from anyone, not just their specified clients.

Attackers make lookup requests using the IP addresses of their targets, then request tons of information, which ends up flooding the targeted servers with huge amounts of DNS information.

Did two wrongs make a bigger wrong?

Spamhaus, a group of related companies based in London and Geneva, was started in 1998 to track and combat email spam and spammers. It maintains a blacklist of Web-hosting companies known to host spammers, and a whitelist of known "clean" Web hosts.

Both lists are used by Internet service providers around the world, and Spamhaus is partly responsible for the huge drop in email spam in recent years.

Some Web-hosting companies have complained they've been unfairly placed on the Spamhaus blacklist. Spammers have launched DDoS attacks against Spamhaus' website and servers. (There's even a "Stophaus" website based in Russia and dedicated to combating what it calls Spamhaus' "underhanded extortion tactics.")

It appears Cyberbunker has both complained and attacked.

Cyberbunker bases its operations in a decommissioned NATO bunker, built to withstand a nuclear war, in the southern Netherlands. The company was founded in 1998 by a group of hackers who proclaimed the "Republic of Cyberbunker," a sovereign state "surrounded by the Netherlands on all borders."

The company pledges not to ask questions about what its clients are up to.

"In most cases we have no idea as to who or where our customers actually are," the Cyberbunker site proclaims. "Customers are allowed to host any content they like, except child porn and anything related to terrorism. Everything else is fine."

Such a policy has attracted some unsavory clients, including the file-sharing site The Pirate Bay, and, according to Spamhaus, the cybercrime gang known as the Russian Business Network. Cyberbunker also claims to have been raided by a Dutch police SWAT team, which apparently found nothing incriminating on the premises.

It was Cyberbunker's alleged hosting of spammers that caused Spamhaus to place both Cyberbunker and its ISP on the Spamhaus blacklist in the fall of 2011.

As a result, Cyberbunker's ISP dropped it as a client, but both the ISP and Cyberbunker posted long manifestos about why Spamhaus was evil.

The issue seems to have lain dormant until March 18, when a false Anonymous campaign called "Operation Stophaus" was proclaimed on the online bulletin board Pastebin.

It listed a litany of complaints against the "tax-circumventing self-declared Internet terrorists" of Spamhaus, then added a variant of the Anonymous "We Are Legion" tagline.

That posting may have been cover for the DDoS attacks that began the same day. In a statement to the New York Times, Sven Olaf Kamphuis, who claimed to speak for Cyberbunker, and whose Google+ page gives his residence as "Republic Cyberbunker," affirmed that the Dutch hosting company was behind the attacks.

"Nobody ever deputized Spamhaus to determine what goes and does not go on the Internet," Kamphuis told the newspaper. "They worked themselves into that position by pretending to fight spam."

It's hard to see how such an attack can be legally justified. The Netherlands has famously lax laws governing the Internet and other digital communications, but odds are Cyberbunker will be facing another SWAT raid very soon.

Fixing a hole

For his blog posting, CloudFlare's Matthew Prince used the headline "The DDoS That Almost Broke the Internet." That's not entirely accurate, since the problems were rather localized.

However, the attack may prompt an overhaul of the DNS system. Prince and others have been vocal about the need to lock down most or all DNS servers so they no longer respond to lookup requests from anyone.

The Stafford girls track and field team has turned an often overlooked part of the sport into one of its strengths.

The Indians have three throwing standouts that can score major points in small or large meets.

That was evident this past Saturday at the Brian Watkins I?95 Invitational at Brooke Point High School.

Stafford?s Sabrina Johnson, India Kithcart and Ebony Pennant took the top three places in the shotput. Pennant and Kithcart went on to finish first and second in the discus, as well.

?In general [throwers] kind of get overlooked because they just specialize and you don?t see their names out there a lot,? Stafford head coach Pete Augrom said. ?But those three girls in the winter got us 24 points in one event.?

Augrom is speaking of the Commonwealth District meet last month in which the trio took the top three honors in the shotput.

Johnson went on to set a school record in the Northwest Region meet with a throw of 37 feet, 11 inches. The junior Colonial Forge transfer placed sixth in Group AAA.

?She had a great finish to her indoor season,? Augrom said.

Augrom said Indians throwing coach Mark Repass is one of the key reasons for the trio?s success. Repass has held his position the past six years. Augrom said he?s helped develop the Indians? uncommon depth.

?A lot of schools will have one or two that are good, but we really have three or four that we feel we can put out there any day and compete for a top five spot at a meet,? Augrom said. ?It?s definitely a place we feel we are strong.?

The Indians will see if their success can continue on Saturday in the Wildcat Invitational at Mountain View. Stafford and Brooke Point will co-host the Spring Qualifying Track and Field Classic at Brooke Point the following weekend.

If you ever saw her on the street, the words glamour, class, and style would immediately come to mind. Her sunny smile would brighten up the most cloudy and gloomy day. Whether wearing a dark winter coat with a white silk scarf or one of her self-knitted sweaters and boots... every eye seems to focus on her the moment she enters any room or even as she walks down the most crowded street. And that includes me.

Cornelia G. Becker is not only a popular international photographer and owner of her own firm, but a published author and poet who creates emotional imagery with her words, both in her native German and in English. But there is something about her you wouldn't notice at first glance, no matter how long that "glance" is. She's also a rape survivor. This also means she's a fighter. She's fought hard to keep that smile shining and that spirit alive. And you can feel it whenever you're around her.

Lucky for me, she still believes in love. It's evident in the poems and breathtaking photographs she shares with the world. I don't know if I could have made it through such an existence in one piece. But for her, it was more than just an existence. She has chosen to live. Whenever I'm with her and see how she travels through life, it's hard to remember the bumpy and treacherous road on which she's had to navigate. Like a delicate butterfly, she lives to find the most beautiful flowers in the ugliest fields.

This, my friends, is what might be considered by the Germans as a Liebeserkl?rung -- a declaration of love. But it's more than that. This is someone for whom I have the utmost respect. Not just because of her physical beautiful, but also for the spiritual power she possesses. What is inside only enhances what is on the outside. There are those moments I discover myself transfixed as I watch her every move. I'm totally mesmerized!

But to my surprise, I find she is unaware of what a treasure she is. The fact is, not enough people have spoken of or shown to her the realities of her value. This, of course, shocks the hell out of me! It's sad how so many are all too happy to pounce on our failures, but are graveyard silent when something positive should be said.

But she carries on. And I watch. I see. I notice the attention she garners and it speaks louder than any words could. She is something special, indeed.

But I'm making up for the oversight of others. I let her know every chance I can, both in words and in deeds, just how special she truly is. I don't want her to ever forget how grateful I am to have her in my life. And it's an honor to do so because I'm a better man just being around her. My respect for her grows with each passing day. And so does my love.

Simulations uncover obstacle to harnessing laser-driven fusion

A once-promising approach for using next-generation, ultra-intense lasers to help deliver commercially viable fusion energy has been brought into serious question by new experimental results and first-of-a-kind simulations of laser-plasma interaction.

Researchers at The Ohio State University are evaluating a two-stage process in which a pellet of fusion fuel is first crushed by lasers on all sides, shrinking the pellet to dozens of times its original size, followed by an ultra-intense burst of laser light to ignite a chain reaction. This two-stage approach is called Fast Ignition, and there are a few variants on the theme. In a recent paper, the Ohio State research group considered the long-discussed possibility of using a hollow cone to maintain a channel for the ultra-intense "ignitor pulse" to focus laser energy on the compressed pellet core. Drawing on both experimental results from studies at the Titan Laser at Lawrence Livermore National Laboratory in California, and massively-parallel computer simulations of the laser-target interaction performed at the Ohio Supercomputer Center (OSC) in Columbus, Ohio, the research team found compelling evidence that the cone-guided approach to Fast Ignition has a serious flaw.

"In the history of fusion research, two-steps-forward and one-step-back stories are a common theme," said Chris Orban, Ph.D., a researcher of the High Energy Density Physics research group at Ohio State and the lead theorist on the project. "But sometimes progress is about seeing what's not going to work, just as much as it is looking forward to the next big idea."

Since the ultra-intense pulse delivers energy to the fuel through relativistic electrons accelerated by the laser interaction, the Ohio State study focused on the coupling of the laser light to electrons and the propagation of those electrons through the cone target. Rather than investigating how the interaction would work on a high-demand, high-cost facility like the National Ignition Facility (NIF), which is also based at Lawrence Livermore National Laboratory and one of the largest scientific operations in the world, the researchers considered experiments just across from NIF at the Titan laser, which is much smaller and easily accessible.

Despite its size and despite having lower total energy, for a brief moment the Titan laser is many thousands of times more intense than NIF, which makes it a decent stand-in as a second-stage ignitor pulse. The OSU-led experimental team focused the Titan pulse on hollow cone targets attached at the tip to copper wires and observed the burst of X-ray photons coming from the copper as a measure of the laser energy to relativistic electron conversion efficiency.

The X-ray signal was much lower from the hollow cones with thicker cone walls. "This was strong evidence to the experimental team that the typical approach to cone-guided Fast Ignition wouldn't work, since thicker cones should be more realistic than thin cones," said Orban. "This is because electrons are free to move around in a dense plasma, much like they do in a normal metal, so the thicker cone target is like a thin cone embedded in a dense plasma."

These intuitions were tested in simulations performed at OSC. Whereas earlier efforts to simulate the laser-target interaction were forced to simplify or shrink the target size in order to make the calculations more feasible, Orban used the LSP code to perform the first-ever, full-scale 2D Particle-In-Cell simulations of the entire laser-target interaction using fully realistic laser fields.

These simulations also included a sophisticated model for the pre-heating of the target from stray laser light ahead of the ultra-intense pulse developed by collaborators at the Flash Center for Computational Science at the University of Chicago.

"We were delighted to help Chris use the FLASH code to provide realistic initial conditions for his Particle-In-Cell simulations," said Don Lamb, director of the Flash Center. "This is an outstanding example of how two groups can collaborate to achieve a scientific result that neither could have achieved alone."

To conduct the simulations, the Ohio State researchers accessed OSC's flagship Oakley Cluster supercomputer system. The HP-built system features 8,300+ Intel Xeon cores and 128 NVIDIA Tesla GPUs. Oakley can achieve 88 teraflops, tech-speak for performing 88 trillion calculations per second, or, with acceleration from the NVIDIA GPUs, a total peak performance of 154 teraflops.

"The simulations pointed to the electric fields building up on the edge of the cone as the key to everything," said Orban. "The thicker the cone is, the further away the cone edge is from the laser, and as a result fewer energetic electrons are deflected forward, which is the crucial issue in making cone-guided Fast Ignition a viable approach."

With both the experiment and the simulations telling the same story, the evidence is compelling that the cone-guided route to Fast Ignition is an unlikely one. While other studies have come to similar conclusions, the group was the first to identify the plasma surrounding the cone as a severe hindrance. Thankfully, there are still many other ideas for successfully igniting the fusion pellet with current or soon-to-be-constructed laser facilities. Any future efforts to spark fusion reactions with these lasers using a two-stage fast-ignition approach must be mindful to consider the neutralizing effect of the free electrons in the dense plasma.

"We could not have completed this project without the Oakley Cluster," Orban noted. "It was the perfect combination of speed and RAM and availability for us. And thanks to the profiling I was able to do, the compute time for our production runs went from two weeks in November 2011 to three or four days as of February 2012."

"Energy and the environment is one of the primary focus areas of the center, and this research fits perfectly into that domain," said Brian Guilfoos, the client and technology support manager for OSC. "Many of our systems were designed and software packages selected to best support the type of computing required by investigators working in fields related to our focus areas."

'Moderate' New England red tide forecasted for 2013

Mar. 25, 2013 ? New England is expected to experience a "moderate" red tide this spring and summer, report NOAA-funded scientists studying the toxic algae that cause blooms in the Gulf of Maine. The "red tide" is caused by an alga Alexandrium fundyense, which produces a toxin that can cause paralytic shellfish poisoning (PSP). Red tide typically occurs annually along some portions of the Gulf of Maine coast. This year's outlook is similar to the 2012 red tide which was also classified as "moderate."

As with the past five forecasts for this region, the 2013 outlook is based on the quantities of the A. fundyense in its cyst (dormant) state detected in Gulf of Maine sediments last fall. These data are combined with a computer model to produce a range of bloom scenarios based on previous years' conditions. This year, the team also used a forecast of toxicity impact developed from 34 years of historical data as part of the 2013 outlook. The 2013 bloom is expected to fall somewhere in the middle in terms of toxicity impact, justifying a "moderate" forecast done by the established method.

"This region is very fortunate to have a long time series of cyst abundance data, toxicity records in shellfish, and long-term measurements of ocean conditions from ships and moored instrumented buoys to develop these two complementary approaches to the seasonal forecast," said Woods Hole Oceanographic Institution (WHOI) biologist Don Anderson.

The forecast team emphasizes the need to consult state and local management agencies for updated harvesting closure information. In order to protect public health, shellfish beds are closed when toxicities rise above a quarantine level, often during the peak harvesting season. Due to effective monitoring by state agencies, there have been no illnesses from legally harvested shellfish in recent years, despite some severe blooms during that time period. There have been, however, several severe poisonings of individuals who ignored closure signs.

"Red tide is a chronic problem throughout the Gulf of Maine, affecting commercial and recreational harvesting interests," said Chris Nash, shellfish program manager for the New Hampshire Department of Environmental Services. "State agencies are responsible for monitoring toxicity levels in shellfish harvest areas and implementing harvest closures when needed. As a state manager, regional-scale, seasonal outlooks help us plan and use limited monitoring resources effectively. Ultimately our goals are to protect public health and give consumers confidence in the quality of the seafood products they purchase from markets and restaurants, and these forecasts are useful in realizing those goals."

Project researchers regularly share their field observations and models with more than 150 coastal resource and fisheries managers in six states as well as federal agencies such as NOAA, the FDA and the EPA. Real-time forecasts are updated on a weekly basis and additional information will be provided on the "Current Status" page of the Northeast PSP website. The National Weather Service is also providing extended hydrological and meteorological outlooks to accompany the bloom forecasts.

"NOAA-funded research has led to the development of seasonal forecasts which aid in monitoring and planning for red tides," said Quay Dortch, program coodinator for NOAA's Ecology and Oceanography of Harmful Algal Blooms (ECOHAB) Program. "These forecasts will be an important part of the Operational HAB Forecasting System NOAA is developing to reduce the impacts of harmful algae."

The forecasting project is a collaboration of investigators from NOAA's National Ocean Service, National Weather Service and National Marine Fisheries Service, WHOI, NCSU, University of Maine, the FDA, Maine Department of Marine Resources, New Hampshire Department of Environmental Services, Massachusetts Division of Marine Fisheries, and the North Atlantic Clam Association. Funding is provided through the NOAA program Prevention, Control and Mitigation of Harmful Algal Blooms (PCMHAB), led by Dennis McGillicuddy (WHOI). Long-term support for Alexandrium studies in the Gulf of Maine is provided by the NOAA NOS NCCOS Center for Sponsored Coastal Ocean Research (CSCOR) and NIEHS and the NSF through the Woods Hole Center for Oceans and Human Health.

Report: Pentagon must do more to prevent assaults

WASHINGTON (AP) ? A new report required by Congress recommends that the Department of Defense assess how well commanding officers handle sexual assault and harassment complaints when reviewing their job performance.

The Institute of Medicine said in the report released Tuesday that military sexual assault appears to be an important factor in the development of post-traumatic stress disorder. It cited previous research indicating that female veterans with a reported history of military sexual trauma were nine times more likely to have PTSD compared to other female veterans.

"Increased efforts by DOD are necessary, and a zero-tolerance approach should be implemented," said the Institute of Medicine, which provides advice concerning health and science to policymakers in the federal government and private sector.

The recommendation about sexual assaults was part of a broad look at the health needs of troops and veterans involved in the wars in Iraq and Afghanistan. Although most of the returning troops have adjusted well to life after deployment, 44 percent have reported some readjustment problems.

The most common overlapping health problems are PTSD, substance abuse, depression and symptoms attributed to traumatic brain injuries.

But the problems seen today are really just the beginning, the report said.

"Previous wars have demonstrated that veterans' needs peak several decades after their war service," the IOM panel said.

To prepare for those costs, the federal government should undertake long-term cost forecasts like those that Congress requires for Social Security and Medicare. It said those forecasts should be conducted annually and publicly released by the Department of Veterans Affairs and confirmed by an independent expert.

The wars in Iraq and Afghanistan resulted in the deployment of about 2.2 million troops as of mid-December, it said. Women have played a central role in the efforts. They make up 14 percent of active-duty troops and nearly 18 percent of National Guard and Reserve personnel. The panel's recommendations often focused on the needs of returning female veterans. It said that recent research indicates that female veterans have a higher risk of developing depression than their male counterparts, though they are less likely to commit suicide.

"For more than a decade, female military service members have been subject to repeat deployments, have endured prolonged separation from families, have served side by side with men, and have been exposed to harsh wartime conditions, including witnessing death and destruction," the report said.

The IOM report also said that the support services it provides to military families tends to focus on married, heterosexual couple and their children. The panel said the military needs to ensure its support services also help single parents, same-sex couples and stepfamilies.

The sub that took Hollywood director James Cameron to the deepest place in the ocean is being donated to science.

On the first anniversary of his 10.9km solo descent of the Mariana Trench, Mr Cameron told the BBC that he was giving the Deepsea Challenger to the Woods Hole Oceanographic Institution, US.

Initially, parts will be used as add-ons for other subs, but the entire system could dive again in the future.

Mr Cameron said he would like to return to the controls himself at some point.

The director said that the ocean was a vast unexplored frontier, but that funding cuts were now jeopardising research.

When Mr Cameron made his dive in 2012, he became the first person to reach the bottom of the Mariana Trench, which is in the western Pacific Ocean, for 50 years - and the only person to have ever made the descent alone.

The only previous manned dive to this deepest spot, called the Challenger Deep, was carried out by US Navy Lt Don Walsh and Swiss oceanographer Jacques Piccard. They took the plunge to the deepest point in the ocean in a bathyscaphe called Trieste in 1960.

It took Mr Cameron just two hours to reach the seafloor. While his bright green submersible moved through the water like a vertical torpedo, the director was squeezed into a tiny chamber that kept him safe from the colossal pressures that exist almost 11km down.

While at the bottom, he spent several hours exploring the seafloor. 3D cameras captured images for a National Geographic film that is being released later this year.

He told the BBC that donating the vessel to the Woods Hole Oceanographic Institution (WHOI) would give the sub "a second phase" of life after the dive. He said that funding was so scarce that plans to take the vehicle on a second series of dives had not been possible.

He explained: "I'd love to keep the Deepsea Challenger continuously operational. But I think that what I'm going for right now is what I call 'potentially operational'. The way to do that is to preserve the hardware 100%, which we'll do, but more importantly to preserve the culture of the engineering.

"My hope is with the sub's home now at Woods Hole, there will be a residential team in place - and they will have the knowledge of how to bring that sub back online."

New thinking

WHOI is one of the world's foremost research centres dedicated to ocean science. It already operates a a number of submersibles including the famous Alvin vehicle.

Initially, Woods Hole will use some of the components in Mr Cameron's sub to supplement its own vessels.

The lights and cameras from Deepsea Challenger will be installed on Nereus, an unmanned underwater vehicle that has also explored the Mariana Trench.

Dr Dave Gallo, director of special projects at WHOI, said the next stage would be to work out what other technology might be of use - and whether the submersible could be used again.

He added: "It is for one person, so you would have to have someone trained to do it - and we are looking very closely at every option."

The filmmaker said more money should be put into ocean science - and that it was essential to explore the biology and the chemistry of the deepsea.

The preliminary science results of the dive, which were released at the American Geophysical Union's Fall Meeting in San Francisco, suggest that the team had found many species that are new to science.

The presented results came from Mr Cameron's dive to the Challenger Deep in the Mariana Trench; an earlier dive to the New Britain Trench, off the coast of Papua New Guinea; and footage and samples taken from landers in the Sirena Deep.

The director told BBC News: "As an absolute minimum, (there were ) over 68 new species - most are bacteria, some are amphipods, and there is possibly a new sea cucumber... and that number may go way up.

"There were also some quite interesting new species of giant amphipods that were 7-8in long when amphipods are normally 0.5-1in in size."

But the explorer said he was surprised there were so little visible signs of life in the Mariana Trench.

"We expected there to be a big biomass in the sediment... but I expected more macrofauna, the things crawling around on the bottom. I had never seen a seafloor as devoid of macrofauna as the Challenger Deep.

"Is it because of the depth? Is it because it is so far from land and there is very little happening in the water column directly above it?"

Further exploration, he said, with the Deepsea Challenger and other robotic vessels would be the only way to find the answers.

Senate Democratic budget extends standoff with GOP

(AP) ? An exhausted Senate gave pre-dawn approval Saturday to a Democratic $3.7 trillion budget for next year that embraces nearly $1 trillion in tax increases over the coming decade but shelters domestic programs targeted for cuts by House Republicans.

While their victory was by a razor-thin 50-49 vote, it allowed Democrats to tout their priorities. Yet it doesn't resolve the deep differences the two parties have over deficits and the size of government.

Joining all Republicans voting no were four Democrats who face re-election next year in potentially difficult races: Sens. Max Baucus of Montana, Mark Begich of Alaska, Kay Hagan of North Carolina and Mark Pryor of Arkansas. Sen. Frank Lautenberg, D-N.J., did not vote.

White House spokesman Jay Carney praised the Senate plan, saying in a statement it "will create jobs and cut the deficit in a balanced way."

While calling on both sides to find common ground, Carney did not hold out much hope for compromise with Republicans. The rival budget passed by the GOP-led House cuts social programs too deeply, he said, and fails "to ask for a single dime of deficit reduction from closing tax loopholes for the wealthy and well-connected."

The Senate vote came after lawmakers labored through the night on scores of symbolic amendments, ranging from voicing support for letting states collect taxes on Internet sales to expressing opposition to requiring photo IDs for voters.

Final approval came at around 5 a.m. EDT, capping an extraordinary 20 hours of votes and debate. As the night wore on, virtually all senators remained in the chamber, a rarity during a normal business day. But at that hour, most had nowhere else to go.

The Senate's budget would shrink annual federal shortfalls over the next decade to nearly $400 billion, raise unspecified taxes by $975 billion and cull modest savings from domestic programs.

In contrast, a rival budget approved by the GOP-run House balances the budget within 10 years without boosting taxes.

That blueprint? by House Budget Committee Chairman Paul Ryan, R-Wis., his party's vice presidential candidate last year ? claims $4 trillion more in savings over the period than Senate Democrats by digging deeply into Medicaid, food stamps and other safety net programs for the needy. It would also transform the Medicare health care program for seniors into a voucher-like system for future recipients.

"We have presented very different visions for how our country should work and who it should work for," said Senate Budget Committee Chairman Patty Murray, D-Wash.

The long debate got testy at times.

As the clock ticked past 1 a.m., Murray asked senators to show respect for colleagues "who may not be able to stand as long as us, or who are elderly." Sen. David Vitter, R-La., shot back that Republicans were not trying to delay anything, and wondered what flights or other appointments would be missed if senators voted until 7 a.m.

The loudest acclaim came toward the end, when senators rose as one to cheer a handful of Senate pages ? high school students ? for their work in the chamber since the morning's opening gavel. Senators then left town for a two-week spring recess.

Congressional budgets are planning documents that leave actual changes in revenues and spending for later legislation, and this was the first the Democratic-run Senate has approved in four years. That lapse is testament to the political and mathematical contortions needed to write fiscal plans in an era of record-breaking deficits, and to the parties' profoundly conflicting views.

Republicans said the Democratic budget wasn't much of an accomplishment. "The only good news is that the fiscal path the Democrats laid out...won't become law," said Senate Republican Leader Mitch McConnell of Kentucky.

"I believe we're in denial about the financial condition of our country," Sen. Jeff Sessions of Alabama, top Republican on the Budget panel, said of Democratic efforts to boost spending on some programs. "Trust me, we've got to have some spending reductions."

Though budget shortfalls have shown signs of easing slightly and temporarily, there is no easy path for the two parties to find compromise ? which the first months of 2013 have amply illustrated.

Already this year, Congress has raised taxes on the rich after narrowly averting tax boosts on virtually everyone else, tolerated $85 billion in automatic spending cuts, temporarily sidestepped a federal default and prevented a potential government shutdown.

By sometime this summer, the government's borrowing limit will have to be extended again ? or a default will be at risk ? and it is unclear what Republicans may demand for providing needed votes. It is also uncertain how the two parties will resolve the differences between their two budgets, something many believe simply won't happen.

Both sides have expressed a desire to reduce federal deficits. But President Barack Obama is demanding a combination of tax increases and spending cuts to do so, while GOP leaders say they won't consider higher revenues but want serious reductions in Medicare and other benefit programs that have rocketed deficits skyward.

Obama plans to release his own 2014 budget next month, an unveiling that will be studied for whether it signals a willingness to engage Republicans in negotiations or play political hardball.

The amendments senators considered during their long day of debate were all nonbinding, but some delivered potent political messages.

They voted in favor of giving states more powers to collect sales taxes on online purchases their citizens make from out-of-state Internet companies, and to endorse the proposed Keystone XL pipeline that is to pump oil from Canada to Texas refineries.

They also voiced support for eliminating the $2,500 annual cap on flexible spending account contributions imposed by Obama's health care overhaul and for charging regular postal rates for mailings by political parties, which currently qualify for the lower prices paid by nonprofits.

In a rebuke to one of the Senate's most conservative members, they overwhelmingly rejected a proposal by Sen. Rand Paul, R-Ky., to cut even deeper than the House GOP budget and eliminate deficits in just five years.

The Democratic budget's $975 billion in new taxes would be matched by an equal amount of spending reductions coming chiefly from health programs, defense and reduced interest payments as deficits get smaller than previously anticipated.

This year's projected deficit of nearly $900 billion would fall to around $700 billion next year and bottom out near $400 billion in 2016 before trending upward again.

Shoehorned into the package is $100 billion for public works projects and other programs aimed at creating jobs.
